Cơ cấu trung ương đầu tiên của Đảng gồm 16 người với Trương Tử Anh làm Đảng trưởng, có trụ sở đặt ở Hà Nội. Trung ương trực tiếp điều hành Xứ bộ Bắc Việt trong khi Trung Việt và Nam Việt có xứ bộ riêng. Ngoài ra Đảng còn có nhân sự hoạt động ở Lào và Cao Miên. Bên cạnh đó, một chi bộ Đảng đặc biệt được thành lập ở Phú Yên được gọi là Chi bộ Đảng-trưởng đặt dưới quyền trực tiếp của Trung ương. Những đảng viên chủ chốt bấy giờ có Nguyễn Tiến Hỷ, Nguyễn Sĩ Dinh, Phan Cảnh Hoàng, Trương Bá Hoành, Đặng Vũ Trứ, Nguyễn Sơn Hải, Tạ Thành Châm, Phan Bá Trọng, Nguyễn Tôn Hoàn, Nguyễn Định Quốc, Đặng Xuân Tiếp, Đặng Vũ Lạc, Ngô Gia Hy, Đặng Văn Sung, Bùi Diễm, Trần Trung Dung, Nguyễn Đình Luyện, Phan Huy Quát, Lê Thăng, Bửu Hiệp, Hà Thúc Ký, Hoàng Xuân Tửu, Dương Thiệu Di. Nguyễn Tôn Hoàn được giao phó làm phát ngôn viên của Đảng và liên lạc viên giữa các Xứ bộ.
